Assess the effect of your compounds on skin explants

Prior to testing compounds or formulations on actual human patients in the context of clinical research in cosmetics or dermatology, human skin explants are now the reference protocols to initially test candidates and get robust evidence of their non-toxicity and efficacy. This method is now the most valuable solution as an excellent alternative to animal testing.

Skin explants are adapted to evaluate toxicity, efficacy, and biocompatibility of your products.

 

Skin explants culture

Skin explant biopsies from healthy donors (from abdominal, face or breast) are managed according to ethical rules and quality procedures and are processed by our technicians using inserts:
• Culture fromJ1 to J7
• Treatment by topical application, intradermal injection or medium stimulation
• Daily medium renewal

Supernatants and skin explants are collected and analysed in our labs to evaluate the toxicity and/or the efficacy of the tested conditions. Many markers of skin pigmentation, hydratation, vascularization and inflammation can be explored for objectivation and comparison of products/formulation.

 

Gene & protein expression

A lot of parameters can be analysed by RT-qPCR or ELISA to evaluate intercellular communication, extracellular matrix components, dermo-epidermal junction, epidermal differentiation

 

Histological processing & analysis

75 stainings & immunostainings, IHC & IF, and protocol development (see our catalog) for global structure or specific markers observations as epidermal differentiation, epidermal cohesion and intercellular exchanges, synthesis and expression of extracellular matrix components, dermal epidermal junction proteins, pigmentation…

Once generated, histological slides are scanned and analysed bymanual/automated methods. Other imaging analyses are also available and can be adapted to match your
needs: area measurement, vessel and cells quantification, epidermal thickness, fluorescence intensity…
